Studies on Insulin Action. 11. the Extracellular Space of Frog Muscle: Demonstration of D-mannitol and Sucrose Entry into Isolated Single Muscle Fibers and Intact Muscles
نویسندگان
چکیده
The distribution of labeled D-mamitol, sucrose, and other widely used extracellular space probe materials was studied in four types of muscles from leopard frogs and bullfrogs as well as in connective tissues and isolated single and multiple muscle fiber preparations from bullfrog semitendinosus muscle. In leopard frog muscle the extracellular space complex (consisting of the extracellular space proper along with connective tissues, small blood vessels, nerves, the sarcolemma and the transverse tubular system), accomrnodates D-mannitol and sucrose to a total concentration of about 9% of that of the external solution. Both D-mannitol and sucrose enter into muscle cells; an equilibrium concentration in the cell water of from 12.5% to as high as 34% of that in the external solution was observed.

Action of Insulin on Sugar Permeability in Rat Diaphragm Muscle
Insulin action upon sugar permeability has been examined in rat diaphragm muscle prepared so that the fibers are either intact or cut. In intact preparations, sucrose and mannitol are largely excluded from the intracellular water while D-galactose, D-xylose, and L-xylose equilibrate in a small fraction of the total cell water; the availability of cell water for D-galactose, D-xylose, and L-xylo...
متن کاملHistopathologic Changes of Extraocular Mus-cle Fibers after Injection of Botulinum Toxin
Background: Use of botulinum toxin is becoming a popular way in treatment of some forms of strabismus. To evaluate the morphological changes in the orbital surface layer muscles of the rabbit eye following a single injection of botulinum toxin. Methods: 10 white Dutch rabbits weighing 2–3 kg received either 10 units (group 1; n=5) or 20 units (group 2; n=5) of botulinum toxin into the superior r...
متن کاملRegulation of cell membrane permeability in skeletal muscle. I. Action of insulin and trypsin on the transport system for sugar.
The mechanism of the stimulatory action of insulin on the transport of sugar across the plasma membrane of skeletal muscle cells was studied by kinetic analysis of the penetration of 3-O-methyl-D-glucose into isolated frog sartorius muscles. As part of this evaluation, the effects of trypsin and insulin were compared. Phospholipase C exerted a much smaller effect on permeability than either of ...
متن کاملContraction and Action Potentials of Frog Heart Muscles Soaked in Sucrose Solution
Isolated auricles or ventricles from the frog continue to contract, either spontaneously or when stimulated, for from 2 to 4 hours after they are placed in isotonic sucrose solution. After the muscles stop contracting in sucrose solution, contractility is partially restored when the muscles are placed in chloride Ringer's. However, contractility is usually not restored if the muscles are placed...
متن کاملThe significance of the action of insulin upon muscle membrane potential.
Two observations placed in juxtaposition may clarify the mechanism of action of insulin. The first is that insulin in contact with intact rat muscle causes net movement of potassium from extracellular fluid into muscle. The second is concerned with the electrical potential difference across the cell membrane of resting muscle which depends in part upon the relative concentrations of potassium o...
